Voici le mode op\u00e9ratoire de la coproculture et <strong>la proc\u00e9dure pratique du d\u00e9roulement de ce test au laboratoire<\/strong>.<\/p>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h1>Coproculture : Technique<\/h1>\n\n\n\n<h2>1. Pr\u00e9l\u00e8vement des selles<\/h2>\n\n\n\n<ul><li>Utiliser un r\u00e9cipient st\u00e9rile (pot \u00e0 selles).<\/li><li>Pr\u00e9lever une petite quantit\u00e9 de selles (\u00e9viter contamination avec urine ou eau).<\/li><li>Si selles liquides : pr\u00e9lever 2\u20135 mL.<\/li><li>Si selles solides : pr\u00e9lever une noix (~2\u20133 g).<\/li><li>Transport rapide au laboratoire (\u2264 2 heures).<\/li><li>Si d\u00e9lai : conserver \u00e0 4\u00b0C ou utiliser un milieu de transport (type Cary-Blair).<\/li><\/ul>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2>2. Examen macroscopique<\/h2>\n\n\n\n<p>Observer :<\/p>\n\n\n\n<ul><li>Couleur (normale, noire, rouge)<\/li><li>Consistance (liquide, p\u00e2teuse, solide)<\/li><li>Pr\u00e9sence de mucus, sang ou parasites visibles<\/li><\/ul>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2>3. Examen microscopique:<\/h2>\n\n\n\n<ul><li>Pr\u00e9paration \u00e0 l\u2019\u00e9tat frais (solution saline)<\/li><li>Recherche de :<ul><li>Leucocytes (infection bact\u00e9rienne)<\/li><li>H\u00e9maties<\/li><li>bact\u00e9ries mobiles : Campylobacter ..<\/li><li>Parasites (\u0153ufs, kystes)<\/li><li>levures<\/li><\/ul><\/li><\/ul>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2>4. Ensemencement des milieux de culture<\/h2>\n\n\n\n<p>\u00c0 l\u2019aide d\u2019une anse st\u00e9rile :<\/p>\n\n\n\n<h3>Milieux classiques utilis\u00e9s :<\/h3>\n\n\n\n<ul><li><strong>G\u00e9lose MacConkey<\/strong><br>\u2192 s\u00e9lectionne les ent\u00e9robact\u00e9ries et diff\u00e9rencie lactose + \/ &#8211;<\/li><li><strong>G\u00e9lose Hektoen<\/strong><br>\u2192 isolement de Salmonella et Shigella<\/li><li><strong>G\u00e9lose SS<br>\u2192 sp\u00e9cifique Salmonella\/Shigella<\/strong><\/li><li><strong>G\u00e9lose XLD<\/strong><br>\u2192 diff\u00e9renciation ent\u00e9robact\u00e9ries pathog\u00e8nes<\/li><li>(optionnel) g\u00e9lose Campylobacter, si suspicion<\/li><\/ul>\n\n\n\n<p>Technique :<\/p>\n\n\n\n<ul><li>Strier en quadrants pour isolement des colonies<\/li><\/ul>\n\n\n\n<p>En pratique on utilise uniquement le milieu sp\u00e9cifique <strong>SS.<\/strong><\/p>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2>5. Incubation<\/h2>\n\n\n\n<ul><li>Temp\u00e9rature : 35\u201337\u00b0C<\/li><li>Dur\u00e9e : 18\u201324 h (jusqu\u2019\u00e0 48 h si n\u00e9cessaire)<\/li><li>Atmosph\u00e8re :<ul><li>A\u00e9robie pour la plupart<\/li><li>Microa\u00e9rophile pour Campylobacter<\/li><\/ul><\/li><\/ul>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2>6. Lecture des cultures<\/h2>\n\n\n\n<p>Observer :<\/p>\n\n\n\n<ul><li>Aspect des colonies (taille, couleur, forme)<\/li><li>Lactose + (rose sur MacConkey) vs lactose \u2013 (incolore)<\/li><li>Production de H\u2082S (colonies noires)<\/li><\/ul>\n\n\n\n<p>Exemples :<\/p>\n\n\n\n<ul><li><strong>Salmonella<\/strong> \u2192 colonies incolores avec centre noir<\/li><li><strong>Shigella<\/strong> \u2192 colonies incolores sans H\u2082S<\/li><li><strong>Escherichia coli<\/strong> \u2192 colonies roses (lactose +)<\/li><\/ul>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2>7. Identification bact\u00e9rienne<\/h2>\n\n\n\n<ul><li>Tests biochimiques :<ul><li><strong>Galerie API 20E<\/strong><\/li><li>TSI, ur\u00e9ase, citrate\u2026<\/li><\/ul><\/li><li>Identification automatis\u00e9e possible (VITEK, MALDI-TOF)<\/li><\/ul>\n\n\n\n<hr class=\"wp-block-separator has-alpha-channel-opacity\"\/>\n\n\n\n<h2>8.
